Phenotypic and environmental data for Lake Ring, Denmark, Daphnia magna population, 1955-2010

This dataset contains fitness-linked life history traits, environmental data, and protein and gene expression data for Daphnia magna. Life history data were measured in common garden experiments. For each clone of the same genotype across all experiments, size at maturity (distance between the head and the base of the tail spine), age at maturity (first time eggs were observed in the brood chamber), fecundity (total number of offspring released summing first and second brood), and mortality were measured. Critical thermal maximum (CTmax) was measured on the experimental animals. Environmental data consisted of Secchi disk depth (water transparency) ; total phosphorous; and total nitrogen for the years 1971–1999; a record of pesticides 1955–2010; temperature records over the past century 80 km from Lake Ring. Organic and carbonate contents of the sediment was estimated using the loss on ignition (LOI) method. Hb protein data consisted of constitutive Hb protein crude content animals reared in normoxic (saturated oxygen level) conditions at two experimental temperatures, 20 and 30°C. Heat shock protein expression was measured in four heat shock proteins (HSP20, HSP60, HSP70 and HSP90). Total RNA, qPCR, mean CT (cycle threshold) value per sample and per protein were collected. Statement
Lineages (genotypes) of Daphnia were resurrected (revived) from biological archives of Lake Ring, a shallow mixed lake in Denmark (550 57´ 51.83´´ N, 9035´46.87´´E) with a known history of anthropogenic impact. The process by which lineages were obtained is as follows: 1. Sediment is sampled using a piston corer of approximately 1.5 m in length with an internal tube diameter of 14 cm 2. The sediment core is separated in incremental layers of 1 cm or less using a flat metal surface 3. Each sediment layer is collected in a separate sampling bag and weighted using a precision scale 4. Each layer of sediment is sieved and dormant Daphnia isolated after visual inspection 5. Under a stereo microscope, each dormant Daphnia encapsulated in an ephippium is opened using microdissection forceps to expose dormant embryos to light 6. Each dormant embryos develops in a fully functional adult from which an isoclonal lineage is established and maintained in the laboratory indefinitely. From a total of 923 dormant Daphnia, 262 lineages were successfully hatched. From these, a total of 40 randomly chosen lineages across the sediment core were used in the experiments and fitness-linked life-history traits measured.
